Laboratory Description

There are two major study interests in our laboratory. The first is interested in the investigation of the immunopathogenesis of influenza A virus infection and establish novel therapeutic strategy to prevent or to treat influenza infection. The second is to investigate cytokines and chemokines in the pathogenesis of autoimmune diseases, including multiple sclerosis and type 1 diabetes.

One of our study in influenza A virus infection demonstrated that melatonin treatment significantly increases the survival of influenza infected mice. This treatment decreases the expression of TNF-α, IL-6 and IFN-γ, and increases the production of IL-10 and TGF-β. The induction of IL-10 by melatonin occurs via the upregulation of IL-27 expression in dendritic cells. Melatonin also significantly inhibits the production of TNF-α in CD8 T cells. Co-treatment of melatonin and ribavirin significantly increases the survival of virus-infected mice compared to ribavirin alone. This study suggests that melatonin possesses a therapeutic potential in influenza-induced pneumonia as an adjuvant treatment with anti-viral drugs. In the study of autoimmune diseases, we are focused on the investigation of the pathogenesis mechanisms in multiple sclerosis and type 1 diabetes and development of novel therapeutic strategies for these diseases. One of our studies is to generate a transgenic NOD mice in which specifically overexpressed an inflammatory CC chemokine decoy receptor D6 on β cells. By analyzing the influence of D6 overexpression in this transgenic mouse, we revealed that inflammatory CC chemokines play a critical role in the development of autoimmune diabetes by promoting the activation and migration of autoreactive lymphocytes. Moreover, we established an animal model of islet transplantation in NOD mice. We are also work in overcome allograft rejection and autoimmune recurrence in a islet transplantation model for the treatment of type 1 diabetes. By using this animal model, we evaluated the protective potential of melatonin and triptolide for prolonging islet graft survival in the therapy of type 1 diabetes. Furthermore, we also evaluated whether co-overexpression of two protective genes, decoy receptor 3 and heme oxygenase-1, in islet can protect islet grafts from autoimmune destruction. However, we found that co-overexpresses two protective genes in islet did not exhibit a protection effect on islet grafts but decrease the survival time of these grafts.
